Electron and Confocal Microscopy LaboratoryIntroduction to the LaboratoryLight microscopy is an invaluable tool in the diagnosis of disease in the pathology
laboratory as well as in the research laboratory. There are times, however,
when the diagnostic features of a disease are beyond the limits of resolution
of traditional light microscopy. The Imaging Laboratory, located within
the Department of Pathology and Laboratory Medicine, offers a variety
of services such as Fluorescence, Confocal and Electron Microscopy, that
go beyond the limits of light microscopy. Fluorescence MicroscopyThe Laboratory is equipped with two Olympus BX51 epifluorescence microscopes each of which has a variety of filters covering numerous fluorophores. Each microscope is equipped with a Spot slider digital camera and corresponding software for digital image acquisition. Electron MicroscopyThe Laboratory is equipped with a JEOL JEM-1230 transmission electron microscope. Transmission Electron Microscopy allows for high resolution ultrastructural analysis of ultrathin sections of biological, particle, and materials specimens. The TEM is equipped with a digital camera and AMT software for digital image acquisition.Confocal Microscopy and Image AnalysisThe Laboratory is equipped with a Leica laser scanning confocal microscope that allows the user to optically section through a specimen from top to bottom. Both upright and inverted microscopes are available. Each has the capacity to collect data simultaneously from up to three fluophores plus a transmitted light image. The laboratory is also equipped to do three-dimensional reconstructions and volume analysis of confocally scanned images.
